Source text - French Zika: la course aux vaccins s'accélère
Une équipe a annoncé le succès de ses tests sur les macaques, d'autres lancent déjà des essais chez l'homme.
**Alors qu'au Brésil, pays le plus touché par l'épidémie due au virus Zika, avec près d'un million et demi de personnes infectées depuis 2015, que des cas sont signalés dans la plupart des pays d'Amérique latine, que les États-Unis viennent de recenser 14 cas de contamination locale -les précédents étant importés de pays où le virus est en circulation-, de bonnes nouvelles commencent à filtrer du côté des laboratoires.
Jeudi, la revue Science annonçait ainsi le succès d'une collaboration entre l'Institut de recherche biomédicale de l'armée américaine (le Walter Reed Army Institute of Research), l'hôpital new-yorkais Beth Israel et la faculté de médecine de Harvard. Ces instituts ont toutes les techniques de biologie moléculaire et de virologie pour mettre au point trois vaccins candidats. Et les essais précliniques, menés sur des souris et des macaques, ont obtenu des résultats encourageants.
«Les singes vaccinés sont protégés contre les souches brésilienne et portoricaine du virus Zika, comme en témoigne l'absence de virus dans le sang, les urines et les sécrétions après une infection», annoncent ainsi les chercheurs. Compte tenu du succès de l'expérience et de l'absence d'effet secondaire, des essais cliniques chez des volontaires devraient être menés d'ici la fin de l'année.
«Ces recherches ont probablement débuté à la fin de l'année dernière. La rapidité avec laquelle ils ont obtenu leurs résultats est incroyable», souligne Étienne Simon-Lorière, virologue à l'Institut Pasteur à Paris. Depuis quelques mois, les recherches sur Zika bénéficient d'un engouement mondial. «De nombreuses équipes se sont mises à travailler sur ce virus, en raison de l'urgence de santé publique mondiale et des importants financements qui lui sont destinés», raconte-t-il, avant de préciser que le nombre de publications sur le virus s'est multiplié de façon exponentielle ces derniers mois.
Gagner un temps précieux
Autre phénomène apparu avec l'épidémie: l'open science (l'ouverture des données scientifiques). «Comme cela a été le cas avec les recherches sur le virus Ebola, certains scientifiques qui travaillent sur Zika ont partagé publiquement leurs résultats, au lieu de passer par le circuit classique de publication, qui peut s'avérer long et fastidieux», poursuit le spécialiste. Ainsi, au début de l'année, l'équipe du Pr David O'Connor (Université du Wisconsin-Madison) a mis en ligne ses résultats dès qu'ils étaient obtenus, avant de les publier dans la prestigieuse revue Nature Communications. Leur découverte -le fait que le macaque est un modèle animal pertinent et proche de l'homme pour l'étude de Zika- a permis à d'autres équipes de gagner un temps précieux.
Cette semaine, la course aux vaccins a pris une autre dimension avec l'annonce, mercredi, par l'Institut américain des allergies et des maladies infectieuses (NIAID) du lancement d'un essai clinique chez 80 volontaires sains âgés de 18 à 35 ans. C'est le second vaccin expérimenté chez l'homme. Une équipe américano-canadienne s'est aussi lancée en juillet.
Le vaccin du NIAID, dont la première dose a été donnée mardi, ne contient pas de virus vivant, mais un brin d'ADN servant de support à la fabrication de l'enveloppe du virus (comme l'un des trois vaccins développé par l'armée américaine). «Cette piste est encourageante mais il n'existe à l'heure actuelle aucun vaccin fonctionnant de cette manière, note toutefois Étienne Simon-Lorière. Sans compter que plusieurs études ont montré que ce type de vaccin peut réagir avec d'autres virus, comme celui de la dengue, en l'aidant à infecter les cellules.»
Selon l'OMS, une dizaine de laboratoires pharmaceutiques dans le monde travaillent actuellement au développement d'un vaccin. Quelle que soit l'issue de ces expériences, le vaccin choisi sera préventif et ne pourra donc pas être administré aux femmes enceintes qui sont les plus à risque.**
Comme redouté, le virus s'installe aux États-Unis
L'arrivée du Zika sur le sol des États-Unis était malheureusement attendue. Cette semaine, les autorités sanitaires de Floride ont confirmé les premiers cas «autochtones», signifiant qu'un moustique a contaminé le malade sur son lieu de vie et non pas lors d'un déplacement à l'étranger. Selon le dernier décompte, 15 personnes ont été infectées dans cet État du sud des États-Unis, dont 12 dans le périmètre très restreint du quartier de Wynwood, à Miami.
Le Centre de contrôle et de prévention des maladies (CDC) a émis pour la première fois de son histoire une restriction de voyage pour raison sanitaire sur le sol américain, en déconseillant aux femmes enceintes de se rendre dans cette partie de la ville. Les mesures pour contrôler la population de moustiques ont échoué, a reconnu le directeur des CDC, Tom Frieden.
«C'est peut-être parce qu'ils sont résistants aux pesticides ou parce qu'ils ont des lieux de ponte difficiles à trouver.» Les espèces de moustiques capables de transmettre Zika (Aedes albopictus et aegypti) sont présentes dans tout le sud des États-Unis et sur une large partie de la moitié est. Arguant de leurs expériences passées avec la dengue et le chikungunya, les autorités ne craignent pourtant pas que les foyers épidémiques ne dépassent l'échelle locale.

“The vaccinated monkeys are protected against the Brazilian and Puerto Rican strains of Zika, as evidenced by the absence of the virus in their blood, urine and secretions post-infection,” the researchers announced. Given the success of the experiment and the absence of side effects, clinical trials in human volunteers should begin by the end of the year.
“These investigations probably started at the end of last year. The quick turnaround for their Zika vaccine results is incredible,” said Stephen Simon Lorière, a virologist at the Institut Pasteur in Paris. In recent months, the research on Zika benefited from the global craze. “There are several teams working on this virus, because of the global public health emergency declared and the substantial funding allocated,” he continued, before adding that the number of publications on the virus had increased exponentially in recent months.
Saving on Precious Time
Another phenomenon appeared along with the epidemic: open science (making scientific research, data and dissemination accessible to all). “As was the case with research on the Ebola virus, some scientists working on Zika shared their results online, instead of going through the conventional publishing circuit, which can be a long and tedious process,” continued the specialist. This week, the race to develop a vaccine took a new turn with the U.S. Institute of Allergy and Infectious Diseases (NIAID) announcement on Wednesday, to launch a clinical trial with 80 healthy volunteers aged between 18 and 35. This is the second vaccine experimented on humans. A US-Canadian team also launched another clinical trial in July.
The first dose of the NIAID’s vaccine that was given on Tuesday did not contain the live virus, but a strand of its DNA was used as a carrier in the manufacture of the virus envelope (much like one of the three vaccines developed by the US Army). “This development is encouraging. However, there is currently no vaccine effective in this manner," noted Stephen Simon-Lorière. "Furthermore, several studies showed that this type of vaccine could react with other viruses, such as dengue, in helping them to infect cells.”
According to WHO, there are about ten pharmaceutical companies in the world working to develop a vaccine. Whatever the outcome of these experiments, the vaccine selected will be preventative and therefore cannot be given to pregnant women, who are the ones most at risk. **
As Feared, The Virus Has Reached the United States
The arrival of Zika on United States soil was unfortunately expected. This week, the Florida Health Authorities confirmed the first-known mosquito-borne cases, meaning that a mosquito contaminated the infected person where they live and not while traveling abroad. At last count, 15 people were infected in this southern State of the U.S., including 12 cases reported in the very restricted perimeter near Miami’s Wynwood neighborhood.
The Center for Disease Control and Prevention (CDC) issued a historic health-related travel warning for the first time ever on American soil, discouraging pregnant women from visiting this part of town. “The measures taken to control the mosquito population failed,” recognized Tom Frieden, the Director of the CDC.
“That might be because they were resistant to pesticides or because their spawning grounds were hard to find.” The mosquito species able to transmit Zika (Aedes albopictus and aegypti) are present throughout the south and a large part of the eastern half of the United States. Citing past experiences with dengue and chikungunya, the authorities have not yet feared that the outbreak will expand beyond local perimeters.
