Amphetamine and Other Psychostimulants

This chapter discusses drugs that are taken for psychostimulation, especially amphetamine and related indirect bioamine agonists but also other substances with different pharmacological properties. (Cocaine, because in the 1980s it became a major American preoccupation, is considered separately in Chapter 5.) Amphetamine-like drugs are used for narcolepsy and attention deficit disorder. It was long believed that their anorectic effects wear off in a matter of weeks, yet a 1992 study found that weight loss was sustained as long as the drugs were taken. They are rarely of value in depressive disorders. Although their manufacture decreased following regulatory legislation in the 1960s and 1970s, amphetamine-like psychostimulants are still produced in huge quantities, both legally and illegally (Table 4-1, Figure 4-1).
Pharmacology and Animal Studies
Acute Effects and Self-Administration
Amphetamine is both a central stimulant and a peripheral sympathomimetic. In animals, it raises systolic and diastolic blood pressure. Low doses reflexly slow the heart rate: higher doses cause tachycardia or cardia arrhythmia. Stimulation of the medullary respiratory center increases the rate and depth of respiration, and excitatory actions on the brainstem, diencephalon, and cerebrum produce increased locomotor activity, sleeplessness, and increased body temperature. Higher or repeated doses cause stereotypic motor behavior (exploration, nose poking, head bobbing) and then tremor, dyskinesias, or catalepsy. Seizures are less frequent than cocaine: in fact, amphetamine raises the threshold for electroshock seizures. Also in contrast to cocaine, amphetamine increases the cerebral metabolic rate for oxygen (CMRO2). As a central nervous system (CNS) stimulant, the D-isomer of amphetamine (dextroamphetamine) is 4 times more potent than the L-isomer. Methamphetamine is more potent than either.

Ce chapitre aborde les drogues qui sont consommées pour leurs effets psychostimulants, et plus particulièrement l'amphétamine et les agonistes indirects des bioamines liées, mais également d'autres substances aux propriétés pharmacologiques différentes (la cocaïne est traitée séparément au chapitre 5, car elle représente une préoccupation majeure des États-Unis depuis les années quatre-vingt). Des drogues aux effets semblables à ceux de l'amphétamine (amphetamine-like) sont utilisées pour le traitement de la narcolepsie et du trouble de déficit de l'attention. On pensait depuis longtemps que leurs effets anorexigènes se dissipaient en quelques semaines, mais une étude réalisée en 1992 a découvert que la perte de poids se poursuivait tant que la drogue était consommée. Effets aigus et autoadministration
L'amphétamine est à la fois un stimulant du système nerveux central (SNC) et un sympathomimétique périphérique. Chez les animaux, elle augmente les pressions artérielles systolique et diastolique. Des doses faibles produisent un ralentissement réflexe de la fréquence cardiaque ; des doses plus élevées provoquent une tachycardie ou une arythmie cardiaque. La stimulation du centre respiratoire bulbaire augmente la fréquence et l'amplitude respiratoires, et les actions excitatrices exercées sur le tronc cérébral, le diencéphale et le cerveau contribuent à augmenter l'activité motrice et la température corporelle et causent des insomnies. L'administration de doses plus fortes ou à répétition entraîne un comportement moteur stéréotypé (exploration, action de pousser à l'aide du museau, balancement de la tête) puis des trémulations, des dyskinésies ou une catalepsie. Les crises convulsives surviennent moins fréquemment qu'avec la cocaïne ; en fait, l'amphétamine élève le seuil de convulsions provoquées par les électrochocs. De même, contrairement à la cocaïne, l'amphétamine augmente la consommation cérébrale en oxygène (CMRO2). Le D-isomère de l'amphétamine (dextroamphétamine) est un stimulant du SNC quatre fois plus puissant que le L-isomère. La méthamphétamine est encore plus puissante que ces deux isomères.

I worked as a freelance English to French translator from 2002 to 2015, when I passed the French certification to become a full-time secondary English teacher as I felt the need to synchronize my work schedule to that of my two children. Now that they are older, and after having worked as a high school teacher for 7 years, I have decided to start again my career as a translator.
