I heard a broadcast on the radio yesterday, in which they were discussing the amount of media attention spent on the Iraq war versus other mundane subjects, such as celebrity news. The theory was that publishing companies are ignoring public opinion polls in deciding what to print. The people or public were referred to several times as "Gran Público".

In English, el gran público is simply "the public," or, in this case, since it is talking about print, you could also say "the reading public" or "readership."

Looks like you're going to be spoilt for choice on this one.
While agreeing with GoodWords and David, I'm under the impression that 'gran público' may have been used, perhaps slightly condescendingly, to refer to what in other times may have been termed 'the masses'.
Although we are talking about books, and thus readership is a more precise term, mainstream audiences is widely used, even in this context (strange, when you don't listen to books, but that's the way it is). More fittingly you could say 'mainstream readers'.

most readers - readers in general/audiences
I think if you refer to books, el gran público in English is still referred to as most readers - big audiences in the case of best sellers, e.g., are huge number of copies sold or huge number of readers. I do agree with Alvaro, that el gran público somewhat describes "the masses" - a way of saying people who are not specially selective.
