We used two different conjugation chemistries, one of which employed the linker SATA. In this conjugation, the succinimidyl ester within the linker SATA chemically reacts with a primary amine in the polypeptide. Generally, with copper-free click chemistry, two approaches can be used. In one, the antibody moiety is modified with azido groups on the primary amines whereas the polypeptide is modified with alkyne. In the second, the antibody moiety is modified with an alkyne (e.g., MFCO or BCN) whereas the polypeptide is modified with azido groups on the primary amines. We also varied the ratio of the antibody moiety to the polypeptide, from 1:8 to 1:32.
