Source text - English Toxicokinetic evaluations of a prototype formulation of aripiprazole IM depot were conducted in single-dose IM irritation and toxicity studies in rats and dogs. Systemic exposures to aripiprazole were dose-related and generally higher in females than in males. Plasma concentrations of aripiprazole were measurable at all doses up to Day 43 in rats (Cmax approximately 190 ng/mL at 1 mg/kg)10 and Day 42 in dogs (Cmax approximately 30 ng/mL, AUC0-28 days approximately 300 ng-days/mL at 100 mg/dog),11 indicating sustained absorption over these periods. Clinically relevant exposures were not readily achieved in rats or dogs due to the markedly greater metabolic clearance of aripiprazole in animals versus humans, and because release of drug from the injection site, as desired, was slow. Plasma concentrations of the pharmacologically active metabolite, BMS-337040, were generally below the lower limit of quantitation in both species, whereas plasma concentrations of BMS-337044 (dehydro-aripiprazole, another active metabolite) were measurable at most time points in dogs, but not in rats.

Source text - English The following procedures will be performed at some or all of your study visits.
Approximately 30 minutes before and one hour after injections your study doctor will assess the injection site (place where the injection was made) for redness, swelling, and firmness of area. You and your study doctor will assess the injection site for pain. With the intramuscular injections, you may experience some discomfort at the injection site which should be temporary and should resolve on its own. This is an expected occurrence with intramuscular injections.
Your eligibility to continue in the study will be evaluated.
Your body weight and waist size will be measured.
A Body Mass Index (BMI) will be calculated using your height and weight.
You will receive a thorough physical examination.
During the first two weeks of phase 3, you will be given a blister card of oral aripiprazole tablets and instructed on how and when to take these tablets.

U ondergaat een grondig medisch onderzoek.
Tijdens de eerste 2 weken van fase 3 krijgt u een een blisterkaart met oraal in te nemen tabletten aripiprazol en krijgt u instructies over hoe en wanneer u deze tabletten dient in te nemen.
